  • Phenotype

Germline-specific RNAi against vang-1 effects C. elegans life span. After vang-1(RNAi) in germline-specific RNAi strain NL2098 a significant increase (13%) of mean life span (14.6 d) in comparison to the control (NL2098 kept on RNAi HT115 bacteria carrying the empty ''feeding''-vector) can be observed (12.9 d).

  • Lifespan comparisons

Double mutant rrf-1(pk1417);vang-1(RNAi) has a lifespan of 14.6 days, while single mutant vang-1(RNAi) has a lifespan of 14.9 days, single mutant rrf-1(pk1417) has a lifespan of 12.9 days and wild type has a lifespan of 12.8 days.

RNA-dependent RNA polymerase Family

Locus: CELE_F26A3.8

Wormbase description: rrf-1 encodes an RNA-directed RNA polymerase (RdRP) homolog required for somatic (but not germline) RNAi; RRF-1 is also required for 'transitive interference'.

Vang-like protein

Locus: CELE_B0410.2

Wormbase description: vang-1 encodes an ortholog of Drosophila STRABISMUS/VAN GOGH; vang-1 enables Wnt-directed planar cell polarity; VANG-1 is required for the fully asymmetrical division of B.a versus B.p cells, though this requirement is quantitatively weak; vang-1 also negatively regulates adult lifespan via the DAF-2/IGFR insulin signaling pathway, and also plays a role in thermal tolerance and response to oxidative stress.
